Biological Background: CD263/TRAIL-R3 Function and Localization
- CD263/TRAIL-R3, encoded by the TNFRSF10C gene, is a tumor necrosis factor receptor superfamily member that lacks an intracellular death domain, classifying it as a decoy receptor for the cytotoxic ligand TRAIL (TNF-related apoptosis-inducing ligand).
- The protein functions solely as a receptor that binds TRAIL, but unlike the functional TRAIL receptors TNFRSF10A and TNFRSF10B, it cannot transduce an apoptotic signal due to the absence of the death domain.
- By competing with the pro-apoptotic TRAIL receptors for ligand binding, CD263/TRAIL-R3 protects cells from TRAIL-induced apoptosis, contributing to the regulation of cell death pathways.
- Also known as DcR1, LIT, TRAILR3, or TRID, this receptor is attached to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor, as evidenced by its keywords and structural features.
- It is a glycoprotein with disulfide bonds and a signal peptide, consistent with its membrane localization and role as a cell surface receptor.
- CD263/TRAIL-R3 is expressed at higher levels in normal tissues such as peripheral blood lymphocytes, spleen, skeletal muscle, placenta, lung, and heart compared to tumor cell lines, suggesting a role in protecting normal cells from TRAIL-mediated cytotoxicity.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the extracellular domain (amino acids 26-235) of human CD263/TRAIL-R3, which includes the TRAIL-binding region; therefore, the antibody is expected to recognize the ligand-binding site if the epitope is conformational.
- Given the GPI-anchored nature of CD263/TRAIL-R3, cell surface detection by flow cytometry (FC) may be optimal. Consider using viable, non-permeabilized cells to preserve membrane integrity and receptor conformation.
- As a decoy receptor, CD263/TRAIL-R3 lacks a cytoplasmic tail; thus, the conjugate allows direct detection without secondary reagents, simplifying the FC workflow.
- Titration of the antibody is recommended for each cell type to determine the optimal staining concentration and minimize background.
- For specificity verification, include appropriate controls: isotype control antibodies and cell lines with known CD263/TRAIL-R3 expression levels, such as peripheral blood lymphocytes (which express the protein) versus tumor cell lines (which often show reduced expression).
