  • CD262/DR5/TRAILR2, encoded by the TNFRSF10B gene, is a member of the tumor necrosis factor receptor superfamily. It is also known as Death receptor 5 (DR5) and TRAIL receptor 2 (TRAIL-R2), serving as a receptor for the cytotoxic ligand TNFSF10/TRAIL. Related references: PMID:10549288
  • Upon TRAIL binding, the receptor recruits the adapter molecule FADD and caspase-8, forming the death-inducing signaling complex (DISC). This triggers caspase-8 proteolytic activation and a caspase cascade leading to apoptosis. DR5 also promotes NF-kappa-B activation and is essential for ER stress-induced apoptosis.
  • The receptor is widely expressed in adult and fetal tissues, with very high levels in tumor cell lines (e.g., HeLaS3, K-562, HL-60) and organs such as heart, liver, pancreas, spleen, thymus, prostate, ovary, and uterus. It is not detectable in brain.
