Biological Background: CD244/2B4 Function and Localization
- CD244/2B4, encoded by the CD244 gene, is a transmembrane receptor belonging to the signaling lymphocytic activation molecule (SLAM) family, also known as SLAMF4.
- It serves as an activating receptor on natural killer (NK) cells, triggering cytotoxicity, IFN-γ production, and granule exocytosis upon binding to its ligand CD48. Related references: PMID:10359122 PMID:11714776 PMID:8376943
- Activating signaling involves association with the adaptor SH2D1A/SAP and kinase FYN, leading to downstream phosphorylation of VAV1, while attenuation in the absence of SH2D1A depends on INPP5D/SHIP1 and phosphatases. Related references: PMID:15713798 PMID:10934222
- CD244 acts as a costimulatory receptor, enhancing signals from other NK cell receptors such as NCR3 and NCR1. Related references: PMID:10741393
- During early NK cell development, CD244 may function as an inhibitory receptor, potentially ensuring self-tolerance of developing NK cells. Related references: PMID:11917118
- On CD8+ T cells, CD244 provides costimulatory signals upon binding to CD48 on neighboring cells, and it inhibits inflammatory responses in dendritic cells (by similarity).
- Subcellularly, CD244 is localized to the cell membrane and is enriched in membrane rafts.
- It is expressed in a variety of tissues including spleen, peripheral blood lymphocytes, lung, liver, testis, and small intestine, and is found on all NK cells, monocytes, basophils, TCRγδ+ T cells, and a subset of CD8+ T cells.
Experimental Guidance and Technical Tips
- Given the immunogen encompasses the extracellular domain (amino acids 22-221), this antibody is predicted to recognize the native receptor on live cells, making it suitable for flow cytometry. Titrate the antibody to determine the optimal concentration for your specific cell type.
- As CD244 is expressed on NK cells, CD8+ T cell subsets, monocytes, and basophils, incorporate appropriate lineage markers (e.g., CD3, CD56, CD8) to accurately gate populations in multicolor panels.
- Include relevant isotype controls and fluorescence minus one (FMO) controls to assess background staining and confirm specificity.
- Validate the antibody in your experimental system by comparing staining with known expression profiles and under relevant biological conditions.
