Biological Background: CD24 Function and Localization

  • CD24 is a signal transducer protein, also known as small cell lung carcinoma cluster 4 antigen.
  • It plays a pivotal role in the differentiation of various cell types; signaling is triggered by lectin-like ligand binding to its carbohydrates and transduced via GPI-anchor-derived second messengers.
  • CD24 modulates B-cell activation responses: it promotes antigen-dependent proliferation of B-cells and prevents their terminal differentiation into antibody-forming cells. Related references: PMID:11313396
  • In association with SIGLEC10, CD24 may be involved in the selective suppression of immune responses to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90.
  • It plays a role in the control of autoimmunity (by similarity).
  • CD24 is localized at the cell membrane and attached via a GPI-anchor.
  • Expressed predominantly on B-cells, but also found on T-cells and various cell lines including erythroleukemia and small cell lung carcinoma lines.
  • The protein undergoes alternative splicing and glycosylation, with a molecular weight of approximately 8 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-80) of human CD24, targeting the extracellular domain accessible for live cell staining in flow cytometry.
  • For FC applications, titrate the antibody to determine optimal working concentration and consider using relevant isotype controls.
  • CD24 is a GPI-anchored surface protein; avoid permeabilization unless intracellular staining is intended, as it may affect surface epitope accessibility.
  • Validate staining in the sample system of interest, as expression may vary across different cell types and conditions.
