Biological Background: Melanotransferrin Function and Localization
- Melanotransferrin is a cell membrane protein also known as melanoma-associated antigen p97 (CD228, MFI2, MTf). It is a member of the transferrin family but selectively binds iron and zinc.
- Functions primarily in cellular iron uptake. Binds a single iron atom per subunit and is internalized before recycling back to the plasma membrane.
- The protein is GPI-anchored to the cell surface, featuring domains for metal binding and transport. Its molecular weight is approximately 80 kDa for the full-length form, with a smaller 32 kDa fragment detected in some contexts.
- Predominantly expressed in melanoma cells and certain fetal tissues; also found in normal liver, epithelium, umbilical cord, placenta, and sweat gland ducts.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation, as indicated by keywords such as Glycoprotein, Phosphoprotein, and Disulfide bond.
- Classified under several functional keywords: Ion transport, Iron transport, and Zinc binding, reflecting its role in metal homeostasis.
- The gene (MELTF) undergoes alternative splicing, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The conjugate is directly compatible with flow cytometry. Titration is recommended to determine the optimal antibody concentration for each experimental system.
- Use appropriate single-stain controls and fluorescence compensation settings to avoid signal spillover when multiplexing.
- For immunogen design, the recombinant antigen spans the ectodomain (amino acids 20–708), targeting the mature, membrane-proximal region. This region is expected to be accessible on live cells.
- Ensure cells are viable and surface protein integrity is maintained; avoid fixation methods that may mask or alter the epitope unless validated.
