Biological Background: CD228/MFI2 Function and Localization

  • Encoded by the MELTF gene (also known as MAP97, MFI2), melanotransferrin is a cell-surface glycoprotein of the transferrin family. Alternative splicing yields isoforms, with the full-length protein at ~80 kDa and a secreted isoform at ~32 kDa.
  • Functions in cellular iron uptake: binds one ferric ion per subunit, internalizes, and recycles back to the plasma membrane; may also transport zinc.
  • Originally identified as melanoma-associated antigen p97, it is predominantly expressed in human melanomas, certain fetal tissues, liver, epithelium, umbilical cord, placenta, and sweat gland ducts.
  • Anchored to the cell membrane via a C-terminal GPI moiety, acting as a lipid-anchored transporter rather than a soluble carrier.
  • Post-translational modifications include disulfide bonds, glycosylation, and phosphorylation, as identified by proteomics.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 20-708, encompassing nearly the full-length extracellular domain; verify reactivity with native and recombinant CD228 in your flow cytometry panel.
  • As a directly conjugated antibody, no secondary reagent is needed; titrate from 0.1–1 µg per million cells to determine optimal staining index.
  • Validate specificity using CD228-silenced cells or a known negative cell line; track viability and doublet exclusion to avoid artifacts from dying cells with high autofluorescence.
  • GPI-anchored proteins can be sensitive to trypsin; prepare single-cell suspensions with gentle enzymatic or mechanical dissociation if working with adherent cell lines.
