Biological Background: CD226/DNAM-1 Function and Localization
- CD226, also known as DNAM-1 (DNAX accessory molecule 1), PTA1, or TLiSA1, is a transmembrane glycoprotein receptor expressed on immune cells such as T-lymphocytes and NK cells.
- It functions as a costimulatory receptor that mediates intercellular adhesion, lymphocyte signaling, cytotoxicity, and cytokine secretion in cytotoxic T-cells and NK cells. Related references: PMID:8673704 PMID:9712030

- Mechanistically, phosphorylation by Src kinases (LYN or FYN) enables recruitment of the adaptor GRB2, leading to activation of VAV1, PI3K, PLCG1, ERK, AKT, and calcium fluxes.
