Biological Background: CD226/DNAM-1 Function and Localization

CD226 (also known as DNAM-1, PTA1, TLiSA1) is a cell surface receptor belonging to the immunoglobulin superfamily. It plays a critical role in the immune system, mediating intercellular adhesion, lymphocyte signaling, and the cytotoxicity and lymphokine secretion of cytotoxic T-cells and NK cells. Upon recognition of target cells like virus-infected or tumor cells, CD226 acts as a costimulatory receptor. Binding to its ligands PVR/CD155 or NECTIN2/CD112 on target cells enhances the cytotoxic activity of NK cells and CTLs, facilitating their killing of these cells. Mechanistically, CD226 is phosphorylated by Src family kinases such as LYN or FYN, enabling binding to the adapter GRB2. This interaction leads to activation of VAV1, PI3K, PLCG1, and downstream kinases ERK and AKT, as well as calcium fluxes.
