Biological Background: CD226/DNAM-1 Function and Localization

  • CD226 (DNAM-1) is a cell surface receptor belonging to the immunoglobulin superfamily, involved in intercellular adhesion, lymphocyte signaling, and cytotoxicity mediated by cytotoxic T-cells and NK cells. Related references: PMID:8673704, PMID:9712030
  • It functions as a costimulatory receptor upon recognition of target cells, binding to its ligands PVR/CD155 or NECTIN2/CD112 on virus-infected or tumor cells, thereby enhancing NK and CTL cytotoxic activity. Related references: PMID:26755705, PMID:31253644, PMID:30591568
  • Upon ligand engagement, CD226 is phosphorylated by Src family kinases such as LYN or FYN, then recruits the adapter GRB2, leading to activation of VAV1, PI3K, PLCG1, and downstream kinases ERK and AKT, as well as calcium fluxes.
  • CD226 is localized to the cell membrane as a single-pass transmembrane protein.
  • It is expressed by peripheral blood T-lymphocytes and plays a role in immune surveillance.
