Biological Background: Interferon-induced transmembrane protein 1 Function and Localization
- IFITM1 (interferon-induced transmembrane protein 1) is a multipass membrane protein and member of the dispanin family, also designated 9-27, CD225, IFI17, LEU13, and DSPA2a. It functions as a potent antiviral factor induced by interferons.
- It restricts the entry of diverse viruses—including influenza A, SARS-CoV-1/2, Ebola, Marburg, Dengue, West Nile, HIV-1, and HCV—by blocking viral fusion with host cell membranes after endocytosis. Related references: PMID:26354436, PMID:33270927
- Specifically, IFITM1 inhibits SARS-CoV-2 spike protein-mediated cell entry and syncytia formation, underscoring its role in coronavirus defense. Related references: PMID:33051876
- Beyond antiviral activity, IFITM1 mediates IFN-γ antiproliferative effects by inhibiting ERK activation or arresting the cell cycle in G1 phase in a p53-dependent manner, and it positively regulates osteoblast differentiation.
- In hepatocytes, IFITM1 cooperates with IFITM2 and IFITM3 to target endocytosed HCV virions to lysosomes for degradation, effectively blocking late-stage HCV entry. Related references: PMID:26354436
- The protein localizes to the cell membrane and lysosome membrane, and is modified by S-palmitoylation and phosphorylation, which are crucial for its membrane association and antiviral efficacy. Its expression is highly elevated in bone and multiple cancer types (colon, cervical, esophageal, ovarian, and glioma).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-80) of human IFITM1, which likely presents extracellular/cytosolic epitopes suitable for antibody generation.
- This antibody is intended for intracellular flow cytometry (FC intra); fixation and permeabilization conditions should be optimized for the cell type of interest.
- Considering IFITM1’s expression pattern, researchers may wish to validate detection in cancer cell lines or interferon-stimulated primary cells.
- Since IFITM1 is a transmembrane protein, denaturing or native lysis conditions for complementary techniques (e.g., Western blot) may require evaluation, although this product is not formally validated for such applications.
