Biological Background: Interleukin-15 receptor subunit alpha (CD215) Function and Localization
- Functions as the high-affinity receptor for interleukin-15 (IL-15), enabling both cis and trans signaling mechanisms essential for immune cell homeostasis and activation. Related references: PMID:8530383
- In neutrophils, CD215/IL-15Rα binds and activates the kinase SYK upon IL-15 stimulation, and is critically required for IL-15-induced phagocytosis in a SYK-dependent manner. Related references: PMID:15123770
- Multiple isoforms are generated through alternative splicing; some variants lack IL-15 binding ability, and expression of different isoforms can alter or interfere with normal signal transduction. Related references: PMID:10480910
- The receptor localizes predominantly to the plasma membrane but is also found on intracellular membranes including the endoplasmic reticulum, Golgi apparatus, cytoplasmic vesicles, and the nucleus, with a secreted soluble form detectable in the extracellular space.
- At the protein level, CD215/IL-15Rα is prominently expressed in neutrophils and shows higher expression in fetal brain structures such as the hippocampus and cerebellum; soluble forms are notably abundant across all brain regions.
- The protein is characterized by structural features including a signal peptide, a sushi domain, a single transmembrane helix, and multiple post-translational modifications such as glycosylation, disulfide bond formation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 31–205), making this product suitable for detecting membrane-bound and possibly soluble receptor forms. Validate in the relevant sample system.
- As a directly conjugated antibody, it is ready for flow cytometry without secondary staining. Consider titrating to determine optimal staining concentrations for your cell type.
- Include appropriate isotype controls and live/dead discrimination to ensure specific detection of CD215-expressing cells. Human peripheral blood neutrophils or activated T cells may serve as positive control populations.
