Biological Background: IL-13Rα1 Function and Localization

  • Interleukin-13 receptor subunit alpha-1 (IL-13Rα1), also designated CD213a1, is encoded by the IL13RA1 gene and belongs to the type I cytokine receptor family. It is also known as cancer/testis antigen 19 (CT19).
  • IL-13Rα1 binds interleukin-13 (IL-13) with low affinity and forms a functional receptor for IL-13 upon heterodimerization with IL-4 receptor alpha (IL-4RA). It additionally acts as an alternate accessory protein for IL-4 signaling, though it cannot substitute for the common gamma chain (IL-2RG) in enhancing IL-2 binding.
  • The receptor is a single-pass transmembrane protein localized predominantly to the cell membrane, featuring extracellular ligand-binding domains, a transmembrane helix, and a cytoplasmic tail involved in signal transduction.
  • Ubiquitously expressed, with highest transcript levels in heart, liver, skeletal muscle, and ovary, and lower levels in brain, lung, and kidney. IL-13Rα1 is also detected on B-cells, T-cells, and endothelial cells.
  • Alternative splicing of the IL13RA1 transcript generates multiple isoforms; the canonical isoform has a predicted molecular weight of approximately 48.8 kDa, while glycosylation and proteolytic processing may yield additional bands around 31-48 kDa.
  • Key post-translational modifications include N-linked glycosylation and disulfide bond formation, which are essential for proper folding and ligand binding. The protein is classified under keywords such as "3D-structure," "Alternative splicing," "Disulfide bond," "Glycoprotein," "Membrane," and "Signal peptide."

Experimental Guidance and Technical Tips

  • This antibody was raised against a recombinant fusion protein spanning amino acids 29–343 of human IL-13Rα1, encompassing the entire extracellular domain. This design targets the native, surface-exposed receptor, making the antibody suitable for flow cytometry applications.
  • As an conjugate, the antibody is compatible with standard blue laser (488 nm) excitation and FITC/GFP emission filters. For multicolor flow cytometry, careful panel design and compensation controls are recommended to minimize spectral overlap.
  • Given the ubiquitous expression of IL-13Rα1, appropriate negative controls (e.g., isotype control, fluorescence-minus-one) are essential to distinguish specific from non-specific binding.
  • The product shows reactivity with human samples; cross-reactivity with other species has not been assessed. Users should validate performance in their specific cell model of interest.
