Biological Background: CD204 Function and Localization
- CD204, also known as macrophage scavenger receptor types I and II (MSR1/SCARA1), is a transmembrane receptor belonging to the class A scavenger receptor family.
- This receptor mediates the binding, endocytosis, and phagocytosis of modified low-density lipoproteins such as acetylated LDL (AcLDL) and oxidized LDL (OxLDL), promoting cholesterol uptake and foam cell formation during atherogenesis.
Related references:
PMID:2251254 | PMID:34252459 - CD204 plays a key role in innate immunity by recognizing and clearing pathogen-associated molecular patterns, bacterial cell wall components, and apoptotic cell debris via polyanionic ligand interactions.
Related references:
PMID:10722588 - Upon stimulation by CpG DNA, CD204 activates LYN kinase, leading to SYK phosphorylation and recruitment to endosomal TLR9, thereby amplifying TLR9 signaling.
Related references:
PMID:29057947 | PMID:40980882 - CD204 localizes primarily to the cell membrane and is classified as a glycoprotein and phosphoprotein, featuring coiled-coil and collagen-like domains.
- Multiple isoforms (I, II, III) are generated by alternative splicing, with expression detected in monocyte-derived macrophages, liver, placenta, and brain.
Experimental Guidance and Technical Tips
- For flow cytometry applications, titrate the antibody to determine optimal staining concentrations using human macrophage cell lines or primary monocytes.
- Include appropriate isotype controls and viability dyes to discriminate live/dead cells and ensure specific staining.
- The immunogen spans the extracellular domain (amino acids 77–451), suggesting that the antibody recognizes surface-expressed CD204; performance may vary with fixation/permeabilization protocols.
- Validate antibody performance in the relevant biological sample system, as experimental conditions can influence staining intensity.
