Biological Background: CD204 Function and Localization
- CD204, also known as macrophage scavenger receptor types I and II (MSR1 or SCARA1), is a transmembrane receptor encoded by the MSR1 gene. It is a member of the scavenger receptor class A family and mediates endocytosis and phagocytosis of a wide range of ligands.
- Recognizes and internalizes modified low-density lipoproteins, including acetylated LDL (AcLDL) and oxidized LDL (OxLDL), thereby promoting cholesterol uptake and contributing to foam cell formation during atherogenesis. Related references: PMID:2251254 PMID:34252459
- Binds a variety of pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), such as bacterial cell wall components and apoptotic cell debris, supporting macrophage clearance in innate immune defense. Related references: PMID:10722588
- Upon stimulation by CpG DNA, mediates activation of LYN kinase, which phosphorylates SYK, subsequently recruiting it to endosomal TLR9 to enhance TLR9 signaling pathways. Related references: PMID:29057947 PMID:40980882
- Three major isoforms (I, II, III) are generated by alternative splicing. Isoforms I and II are widely expressed in the liver, placenta, brain, and monocyte-derived macrophages, while isoform II lacks the ability to internalize acetylated LDL.
- Located on the cell membrane with a signal-anchor transmembrane domain; structurally features collagen-like, coiled-coil, and cysteine-rich domains, and undergoes post-translational modifications including glycosylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 77‑451 of human CD204, which includes extracellular domains; the resulting monoclonal antibody likely targets an extracellular epitope, making it suitable for live-cell flow cytometry.
- For flow cytometry (FC) with this conjugate, consider titrating the antibody to determine optimal concentration using relevant human macrophage or monocyte cell lines (e.g., THP‑1, U937) and appropriate isotype controls.
- Validate staining specificity by comparing signal to cells not expressing CD204 or by pre‑blocking with an unconjugated anti‑CD204 antibody; note that CD204 expression can be modulated by inflammatory stimuli.
