Biological Background: CD172b Function and Localization
- Signal-regulatory protein beta-1 (SIRPB1), also known as CD172b, is a cell surface receptor of the immunoglobulin superfamily (IgSF) containing immunoglobulin domains, a transmembrane helix, and a signaling cytoplasmic tail.
- It negatively regulates receptor tyrosine kinase-coupled signaling processes and participates in the recruitment of tyrosine kinase SYK.
- When associated with the adaptor TYROBP, SIRPB1 triggers activation of myeloid cells, playing an immunomodulatory role. Related references: PMID:10604985
- SIRPB1 localizes to the cell membrane and is primarily expressed in monocytes and dendritic cells.
- The protein exists in alternatively spliced isoforms, which may modulate its function; it contains disulfide bonds and is N-glycosylated.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 30–371 of human CD172b, corresponding to the extracellular region, which favors recognition of native cell surface receptor in flow cytometry.
- This -conjugated antibody is validated for direct immunofluorescence staining and flow cytometry; consider titrating the optimal concentration for your specific sample.
- Include appropriate controls such as isotype-matched negative control antibody and, if possible, a known positive cell type (e.g., human monocytes) to validate staining specificity.
