Biological Background: CD172a/SIRPα (Tyrosine-protein phosphatase non-receptor type substrate 1) Function and Localization
- Also known as BIT, MFR, SHPS-1, MYD-1, and p84, CD172a/SIRPα is an immunoglobulin-like cell surface receptor that binds CD47 with high affinity.
- Acts as a docking protein, recruiting the tyrosine phosphatases PTPN6 and PTPN11 to the plasma membrane upon ligand engagement, thereby modulating downstream signaling.
- Functions as an inhibitory receptor, negatively regulating phagocytosis, mast cell activation, and dendritic cell maturation; CD47 binding prevents dendritic cell cytokine production.
- Serves as a receptor for thrombospondin-1 (THBS1), triggering phosphorylation and NADPH oxidase-derived ROS production in non-phagocytic cells. Related references: PMID:24511121
- Plays a role in neuronal adhesion, neurite outgrowth, and synaptic function, supporting cerebellar neuron attachment and glial cell interaction (by similarity).
