Biological Background: CD170/Siglec-5 Function and Localization

  • CD170/Siglec-5 is a sialic acid-binding immunoglobulin-like lectin that functions as a putative adhesion molecule, mediating sialic-acid dependent cell binding.
  • Binds both alpha-2,3- and alpha-2,6-linked sialic acids with similar affinity; however, the sialic acid recognition site may be masked by cis interactions on the same cell surface.
  • Belongs to the CD33-related Siglec family, which includes other immune receptors like CD33 (Siglec-3) and Siglec-6, and is also known as CD33 antigen-like 2 (CD33L2).
  • Localized to the cell membrane as a type I transmembrane protein, containing an N-terminal signal peptide, extracellular immunoglobulin-like domains, and a cytoplasmic tail.
  • Predominantly expressed on monocytic and myeloid lineage cells, including monocytes and neutrophils, and found in peripheral blood leukocytes, spleen, bone marrow, and at lower levels in lymph node, lung, appendix, and pancreas.
  • Contains key structural features such as disulfide bonds, glycosylation sites, and a transmembrane helix; has a predicted molecular weight of approximately 61 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular region (aa 17-434), which may target epitopes involved in sialic acid binding; consider using sialidase-treated cells as a negative control to assess specificity.
  • For flow cytometry, optimize antibody titration with relevant cell types (e.g., human peripheral blood mononuclear cells or isolated neutrophils) and include appropriate isotype controls.
  • Conjugated with , a bright red fluorescent dye, suitable for multi-color panels; verify compatibility with other fluorophores and instrument lasers (excitation ~650 nm, emission ~668 nm).
  • Perform live-cell staining as Siglec-5 is a surface membrane protein; avoid permeabilization buffers that may disrupt epitope accessibility.
