Biological Background: CD162/PSGL-1 Function and Localization
- P-selectin glycoprotein ligand 1 (PSGL-1), also known as CD162 or selectin P ligand, is encoded by the SELPLG gene. It is a SLe(x)-type proteoglycan that functions as a key adhesion molecule in the immune system.
- PSGL-1 mediates calcium-dependent interactions with E-, P-, and L-selectins, enabling the tethering and rolling of leukocytes on endothelial cells, which is the initial step in leukocyte extravasation during inflammation.
- It plays a critical role in the early leukocyte capture and is essential for directing leukocytes to sites of inflammation or injury.
- Besides its role in inflammation, PSGL-1 serves as a receptor for enterovirus 71, highlighting its involvement in host-pathogen interactions.
- Subcellularly, PSGL-1 is a transmembrane protein located on the cell surface membrane.
- It is expressed predominantly on hematopoietic cells, including neutrophils, monocytes, and most lymphocytes.
- Post-translational modifications include tyrosine sulfation, O-linked sialylation, and glycosylation, which are critical for its selectin-binding activity. The protein also undergoes alternative splicing and phosphorylation.
Experimental Guidance and Technical Tips
- Since this antibody is directly conjugated to and raised against the extracellular domain (aa 42-295), it is suitable for live-cell flow cytometry staining without a secondary antibody.
- Titrate the antibody to determine the optimal concentration for FC; use an isotype-matched control to assess non-specific binding.
- Validate performance in known PSGL-1-expressing cells such as human neutrophils, monocytes, or lymphocyte subsets.
- Consider using this antibody in combination with other surface markers for detailed immunophenotyping of leukocyte populations.
