Biological Background: CD161 (KLRB1) Function and Localization

  • CD161 (KLRB1) is a killer cell lectin-like receptor subfamily B member 1, also known as NKR-P1A, CLEC5B, and NKRP1A.
  • It functions as an inhibitory receptor on natural killer (NK) cells, suppressing cytotoxicity.
  • Activation stimulates acid sphingomyelinase/SMPD1, elevating intracellular ceramide levels, and also activates AKT1/PKB and RPS6KA1/RSK1 kinases.
  • Receptor engagement enhances T-cell proliferation induced by anti-CD3 stimulation.
  • The extracellular C-type lectin domain binds the Gal-alpha(1,3)Gal and N-acetyllactosamine carbohydrate epitopes, as well as the endogenous ligand CLEC2D/LLT1.
  • Binding to CLEC2D/LLT1 inhibits NK cell-mediated cytotoxicity and interferon-gamma secretion in target cells.
  • CD161 is a transmembrane glycoprotein containing a signal-anchor, a transmembrane helix, and a disulfide bond.
