Biological Background: CD157/BST-1 Function and Localization
- ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 2, also known as CD157 or BST-1, catalyzes the synthesis of cyclic ADP-ribose (cADPR) from NAD+ and its hydrolysis to ADP-ribose (ADPR). cADPR is a second messenger that mobilizes intracellular calcium. Related references: PMID:7805847
- May be involved in pre-B-cell growth, suggesting a role in immune cell development.
- Localized to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor.
- Expressed in various tissues, including placenta, lung, liver, and kidney.
- Subject to alternative splicing, producing multiple isoforms.
- Post-translational modifications include glycosylation, a disulfide bond, and a GPI-anchor; the protein is a lipoprotein.
Experimental Guidance and Technical Tips
- The immunogen comprises a recombinant fusion protein spanning amino acids 29-292 of human CD157/BST-1, which corresponds to the extracellular domain; this design may support recognition of native cell surface antigen in flow cytometry.
- For flow cytometry, consider performing antibody titration to determine the optimal concentration, and use appropriate live-cell staining protocols with relevant isotype controls.
- Validate this antibody in a human cell line known to express CD157/BST-1, and confirm specificity by comparing staining profiles with appropriate negative controls or knockout models.
- If simultaneous detection of multiple markers is required, verify compatibility with your panel and compensation setup.
