Biological Background: CD150/SLAM Function and Localization

  • SLAMF1 (CD150/SLAM) is a member of the SLAM family of immunoglobulin receptors that acts as a self-ligand, modulating the activation and differentiation of various immune cells (T-cells, B-cells, dendritic cells, macrophages) and interconnecting innate and adaptive immunity.
  • In T-cells, SLAMF1 signaling proceeds via two pathways: one dependent on the adaptor SH2D1A/SAP (which recruits FYN kinase) and another mediated by PTPN11 (SHP-2). Related references: PMID:11806999, PMID:12458214
  • In B-cells, SLAMF1 activates ERK signaling (SYK- and INPP5D-dependent) as well as Akt, p38 MAPK, and JNK1/2 cascades. Related references: PMID:20231852

  • SLAMF1 is a membrane glycoprotein with disulfide bonds and phosphorylation sites; it localizes primarily at the cell membrane and can be secreted.
