Biological Background: CD150/SLAM Function and Localization
- SLAMF1 (CD150/SLAM) is a member of the SLAM family of immunoglobulin receptors that acts as a self-ligand, modulating the activation and differentiation of various immune cells (T-cells, B-cells, dendritic cells, macrophages) and interconnecting innate and adaptive immunity.
- In T-cells, SLAMF1 signaling proceeds via two pathways: one dependent on the adaptor SH2D1A/SAP (which recruits FYN kinase) and another mediated by PTPN11 (SHP-2). Related references: PMID:11806999, PMID:12458214
- In B-cells, SLAMF1 activates ERK signaling (SYK- and INPP5D-dependent) as well as Akt, p38 MAPK, and JNK1/2 cascades. Related references: PMID:20231852
- SLAMF1 also functions as a host cell receptor for Measles virus entry, including its isoform 4.
- SLAMF1 is a membrane glycoprotein with disulfide bonds and phosphorylation sites; it localizes primarily at the cell membrane and can be secreted. It is constitutively expressed on memory T-cells and rapidly induced on naive T-cells, with alternative isoforms in B-cells, dendritic cells, and tumors of the central nervous system.
