Biological Background: CD150/SLAM Function and Localization

  • Signaling lymphocytic activation molecule (SLAM, also known as CD150 or CDw150) is a self-ligand receptor of the SLAM family, encoded by the SLAMF1 gene. It exists in multiple isoforms, including a membrane-bound form and a secreted variant.
  • SLAM modulates the activation and differentiation of a wide variety of immune cells, bridging innate and adaptive immunity. Its signaling is differentially regulated by adaptors SH2D1A/SAP or SH2D1B/EAT-2, leading to distinct outcomes in T and B lymphocytes. Related references: PMID:11806999 PMID:12458214
  • In T cells, SLAM promotes IL-2-independent proliferation and IFN-gamma production. It enhances TCR-induced IL-4 secretion by CD4+ cells and supports T follicular helper cell function in germinal centers.
  • In B cells, SLAM activates the ERK and Akt pathways via SYK and INPP5D, and induces p38 MAPK, JNK1, and JNK2 signaling, contributing to humoral immune response regulation. Related references: PMID:20231852
  • The protein is primarily located at the cell membrane but can also be secreted. It contains one immunoglobulin domain and undergoes phosphorylation, glycosylation, and disulfide bond formation.
  • Constitutively expressed on peripheral blood memory T cells, immature thymocytes, and a subset of B cells, SLAM is rapidly upregulated upon naive T-cell activation. Isoform 4 is found in dendritic cells, macrophages, and certain tumors. Related references: PMID:7617038 PMID:9091591 PMID:25710480
  • SLAM serves as a receptor for measles virus and plays a role in innate responses to gram-negative bacteria by recognizing bacterial surface proteins OmpC/OmpF, triggering phagosome maturation and NOX2 activity. Related references: PMID:20818396

Experimental Guidance and Technical Tips

  • The immunogen spans the extracellular domain (aa 21-237), making the antibody suitable for detecting surface SLAM on live unfixed cells.
  • For flow cytometry, titrate the antibody and include appropriate isotype controls to establish optimal staining concentrations for your specific cell system.
  • Validate the antibody on known SLAM-positive and -negative cell lines or primary immune cells under activation conditions (e.g., stimulated PBMCs).
  • Ensure that your flow cytometer is equipped with a violet laser (405 nm) for detection; compensation may be required in multicolor panels.
