Biological Background: Thrombomodulin Function and Localization

  • Thrombomodulin (CD141/BDCA-3, also known as fetomodulin) is a transmembrane receptor abundantly expressed on vascular endothelial cells, functioning in hemostasis, coagulation, fibrinolysis, inflammation, and angiogenesis. Related references: PMID:10761923
  • It acts as a cofactor for thrombin-mediated activation of protein C on the endothelial surface, initiating the anticoagulant pathway. Related references: PMID:29323190, PMID:33836597, PMID:9395524
  • Thrombomodulin accelerates activation of plasma carboxypeptidase B2 (CPB2), which cleaves C-terminal basic amino acids from kinins and anaphylatoxins, contributing to fibrinolysis inhibition. Related references: PMID:26663133

Experimental Guidance and Technical Tips

  • This conjugate is intended for direct immunofluorescence staining and flow cytometric detection of human CD141/Thrombomodulin on the surface of live or fixed cells.
  • Optimal antibody concentration should be determined by titration in the relevant experimental system; a starting range of 0.5–5 µg/mL is often used for cell surface staining.
  • Use appropriate isotype control and viability dye to discriminate live cells and minimize background.
  • For sample preparation, avoid over-digestion with trypsin as thrombomodulin is trypsin-sensitive; consider using non-enzymatic dissociation reagents.
  • CD141 is highly expressed on vascular endothelial cells; consider using primary endothelial cell cultures or peripheral blood endothelial progenitor cells for positive control.
  • Note that the antibody recognizes an extracellular epitope; permeabilization is not required for surface staining but may be employed for intracellular pools if needed after fixation.
