Biological Background: CD11a (Integrin alpha-L) Function and Localization
- CD11a, also known as integrin alpha-L or LFA-1 alpha chain, is encoded by the ITGAL gene and serves as a leukocyte-specific adhesion molecule.
- Paired with integrin beta-2 (ITGB2), it forms the LFA-1 heterodimer, which acts as a receptor for intercellular adhesion molecules (ICAM1, ICAM2, ICAM3, ICAM4) to mediate leukocyte adhesion and immune synapse formation. Related references: PMID:10846180 PMID:12526797 PMID:1676048
- LFA-1 engagement triggers downstream signaling critical for leukocyte-endothelial transmigration, T-cell cytotoxicity, and natural killer cell function. Related references: PMID:11812992 PMID:15356110
- It also acts as a receptor for non-classical ligands, including F11R, secreted ISG15, and neuronal ICAM5, expanding its role beyond classical immune adhesion. Related references: PMID:15528364 PMID:29100055 PMID:10741396
- Subcellularly, CD11a localizes primarily to the plasma membrane and enriches in membrane rafts upon activation.
- Tissue expression is mainly restricted to leukocytes, consistent with its immune-restricted functions.
- The protein undergoes N-glycosylation and alternative splicing, resulting in observed molecular weight variants (118/129/134 kDa) depending on glycosylation state.
- Key biological keywords include calcium/magnesium-dependent adhesion, host-virus interaction, phagocytosis, and phosphoprotein signaling.
Experimental Guidance and Technical Tips
- This -conjugated antibody is intended for flow cytometry on human leukocyte samples. To determine optimal staining concentration, perform a titration series in your assay system.
- Given widespread leukocyte expression of CD11a, consider using appropriate isotype controls and Fc receptor blocking to reduce non-specific binding.
- The immunogen (aa 26-340) spans the extracellular ligand-binding domain, suggesting suitability for detecting native surface CD11a; however, fixation and permeabilization may alter epitope availability.
- When co-staining with other leukocyte markers, verify fluorochrome compatibility and include single-color compensation controls.
