Biological Background: CD119/IFNGR1 Function and Localization

  • Interferon gamma receptor 1 (IFNGR1, CD119) is a cell surface receptor subunit that mediates crucial immune responses against pathogens and tumors.

  • The intracellular domain of IFNGR1 undergoes conformational changes that allow association with JAK1 and JAK2 kinases; activated JAK1 phosphorylates IFNGR1 to create a docking site for STAT1.   • Phosphorylated IFNGR1 also provides a binding site for SOCS1, which competes with STAT1 to negatively regulate the JAK-STAT signaling cascade.
