Biological Background: CD119/IFNGR1 Function and Localization
- IFNGR1 (CD119) is the alpha chain of the interferon gamma receptor, a member of the immunoglobulin superfamily.
- It binds interferon gamma and, together with IFNGR2, assembles into the functional receptor complex that is essential for antimicrobial, antiviral, and antitumor immune responses.
Related references: PMID:20015550, PMID:10986460, PMID:2971451, PMID:7615558 - Ligand binding triggers JAK1/2 kinase recruitment, leading to phosphorylation of IFNGR1 and the subsequent docking and activation of STAT1. Phosphorylated STAT1 dimerizes and translocates to the nucleus to induce transcription of target genes.
Related references: PMID:28883123 - Phosphorylated IFNGR1 also serves as a docking site for SOCS1, which negatively regulates signaling by competing with STAT1.
- The receptor localizes to the cell membrane and features an immunoglobulin-like domain, disulfide bonds, N‑glycosylation, and undergoes phosphorylation and ubiquitination.
- The full‑length protein has a molecular weight of ~54 kDa; a 21 kDa fragment may arise from alternative splicing or proteolytic cleavage.
Experimental Guidance and Technical Tips
- The immunogen comprises the extracellular domain (amino acids 18–245), so the antibody targets the ligand‑binding region.
- For flow cytometry, perform a titration to determine the optimal staining concentration. Use an appropriate isotype control and include Fc receptor blocking when working with FcR‑expressing cells.
- Confirm reactivity with known IFNGR1‑expressing cell lines or primary cells (e.g., T lymphocytes, macrophages), and consider up‑regulation by IFN‑gamma treatment.
- The direct conjugate allows single‑step detection; avoid using fluorophore‑matched secondary antibodies.
