Biological Background: CD117/c-Kit Function and Localization
- CD117/c-Kit is a receptor tyrosine kinase encoded by the KIT gene, also known as mast/stem cell growth factor receptor, SCFR, or Piebald trait protein (PBT).
- It serves as the cell-surface receptor for the cytokine KITLG/SCF, playing essential roles in cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration, and melanogenesis.
- Upon ligand binding, c-Kit activates multiple downstream signaling cascades, including PI3K-AKT, RAS-MAPK, and JAK-STAT pathways, through phosphorylation of adaptors like PIK3R1, GRB2, and STAT proteins.
- The protein localizes to the cell membrane and can also be found in the cytoplasm, consistent with its role as a transmembrane receptor that undergoes internalization.
- In human testis, CD117 is expressed in spermatogonia and interstitial Leydig cells, and is maintained in ejaculated spermatozoa, suggesting a role in spermatogenesis. Related references: PMID:20601678
- c-Kit is classified as a proto-oncogene, and mutations or aberrant activation are associated with various cancers; it belongs to the type III receptor tyrosine kinase family and contains immunoglobulin-like domains in its extracellular region.
- Post-translational modifications include glycosylation, disulfide bond formation, and ubiquitination, which regulate its stability and signaling.
- The full-length human c-Kit protein has a predicted molecular weight of approximately 110 kDa, in agreement with the observed weight in the product specifications.
Experimental Guidance and Technical Tips
- As this antibody is designed for flow cytometry (FC), it is suitable for surface staining of live cells, targeting the extracellular domain (aa 26-520) of human CD117.
- For optimal performance, titrate the antibody to determine the appropriate concentration for your specific cell type and experimental conditions.
- When analyzing hematopoietic stem cells or mast cells, consider co-staining with lineage markers to gate CD117-positive populations.
- Given the receptor’s internalization upon ligand binding, avoid prolonged incubation with KITLG/SCF during experimental setups that require surface staining.
- Although validated for human, consider potential cross-reactivity with other species based on sequence homology; if needed, verify using the appropriate model system.
