Biological Background: CD101/IGSF2 Function and Localization
- CD101 (IGSF2, V7, EWI-101) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily, characterized by extracellular immunoglobulin-like domains and a cytoplasmic tail.
- It functions as a negative regulator of T-cell activation: inhibits CD3-induced proliferation, downregulates IL2RA (CD25) expression, and reduces IL-2 secretion.
- Mechanistically, CD101 suppresses tyrosine kinases, PLCG1 phosphorylation, and calcium flux, thereby blocking NFAT nuclear translocation essential for T-cell activation.
- The protein is expressed on the plasma membrane of activated T-cells, monocytes, granulocytes, and cutaneous dendritic cells, with tissue presence in lung, thymus, and small intestine.
- CD101 is also detected in some leukemic cells, notably CD4+ CD56+ blastic tumor cells, and in Langerhans cells from Langerhans cell histiocytosis patients.

- For IF/ICC, try paraformaldehyde fixation with and without permeabilization to optimize membrane-localized staining; surface-only detection may avoid background from intracellular transit forms.
- In flow cytometry, include proper isotype controls and validate on human sample types known to express CD101, such as activated T-cells or dendritic cells.
- While not listed as a validated application, the antibody could theoretically work in western blotting (predicted molecular weight ~115 kDa); any use for WB should be independently verified by the user.
