Biological Background: Calreticulin Function and Localization
- Calreticulin is a calcium-binding chaperone that promotes protein folding and quality control in the endoplasmic reticulum (ER) via the calreticulin/calnexin cycle. Related references: PMID:7876246
- It transiently interacts with monoglucosylated glycoproteins to ensure proper folding and assembly. Related references: PMID:7876246
- Calreticulin binds the DNA-binding domain of the glucocorticoid receptor (NR3C1) and mediates its nuclear export. Related references: PMID:11149926
- It is involved in maternal gene expression regulation and may participate in oocyte maturation by regulating calcium homeostasis (By similarity).
- The protein is localized primarily in the endoplasmic reticulum lumen but also found in the cytosol, secreted to the extracellular space, on the cell surface, and in cytoplasmic vesicles.
- Calreticulin undergoes post-translational modifications including acetylation and hydroxylation, contains a disulfide bond, and binds zinc.
- Alternative names include CRP55, Calregulin, ERp60, HACBP, and grp60; its molecular weight is approximately 48 kDa.
- In oocytes, it is stored in cortical granules and exocytosed during the cortical reaction, potentially contributing to the block to polyspermy (By similarity).
Experimental Guidance and Technical Tips
- This antibody targets a C-terminal fragment (aa 335–417) and may be suitable for intracellular flow cytometry after appropriate fixation and permeabilization.
- Optimize staining protocols for your specific cell type; calreticulin’s ER localization requires effective permeabilization to access the epitope.
- Include positive control cells known to express calreticulin, such as human lymphocytes or cell lines.
