Biological Background: Calreticulin Function and Localization

  • Calreticulin is a highly conserved calcium-binding chaperone also known as CRP55, Calregulin, ERp60, HACBP, or grp60. It is the product of the CALR gene in humans (Gene ID: 811).
  • In the endoplasmic reticulum, calreticulin serves as a lectin-like chaperone that promotes the folding, oligomeric assembly, and quality control of nascent glycoproteins via the calreticulin/calnexin cycle. It interacts transiently with almost all monoglucosylated glycoproteins synthesized in the ER. Related references: PMID:7876246
  • Beyond the ER, calreticulin interacts with the DNA-binding domain of the nuclear receptor NR3C1 and mediates its nuclear export, linking the protein to transcriptional regulation. Related references: PMID:11149926
  • Calreticulin is implicated in reproductive biology, where it participates in maternal gene expression regulation and may contribute to oocyte maturation through calcium homeostasis. In non-activated oocytes, it is present in cortical granules and is exocytosed during the cortical reaction to help prevent polyspermy.
  • Subcellularly, calreticulin is predominantly found in the endoplasmic reticulum lumen, but it can also localize to the cytosol, extracellular space, cell surface, sarcoplasmic reticulum, and secretory vesicles such as cortical and cytolytic granules.
  • The protein undergoes various post-translational modifications including acetylation, hydroxylation, and disulfide bond formation. It features a signal peptide, repetitive motifs, and is categorized under multiple keywords: chaperone, calcium, metal-binding (zinc), glycoprotein, lectin, and others.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 335–417 of human calreticulin, a C‑terminal region distinct from the N‑terminal signal peptide and chaperone domains. This design may allow detection of total calreticulin regardless of ER‑retention or secretion status.
  • Since calreticulin resides mainly in the ER lumen, intracellular flow cytometry (FC intra) requires cell fixation and permeabilization. Optimize permeabilization conditions (e.g., using saponin or methanol) to ensure antibody access while preserving epitope reactivity.
  • is a far‑red fluorophore that enables multiplexing with other intracellular markers while minimizing spectral overlap. Titrate the antibody to determine the optimal signal‑to‑noise ratio for your specific experimental system.
  • Consider using this antibody to investigate ER stress, protein quality control dynamics, or calreticulin translocation events in human cell models. As with any intracellular staining, include appropriate controls (unstimulated cells, isotype controls, and fluorescence minus one) to validate specificity.
