Biological Background: BCL6 Function and Localization
- BCL6 (B-cell lymphoma 6) is a transcriptional repressor with a BTB/POZ domain and zinc finger motifs, also known as BCL5, LAZ3, ZBTB27, and ZNF51.
- Primarily required for germinal center (GC) formation and antibody affinity maturation, BCL6 represses genes involved in differentiation, inflammation, apoptosis, and cell cycle control in GC B-cells.
- Functions by binding directly to the DNA sequence 5'-TTCCTAGAA-3' or indirectly by repressing transcription factors, and recruits corepressors and histone deacetylases to silence target genes.
- Allows rapid proliferation of GC B-cells and tolerates DNA breaks during immunoglobulin class switching and somatic hypermutation without inducing p53-dependent apoptosis.
- In follicular helper CD4+ T-cells (TFH), BCL6 promotes TFH gene expression while inhibiting differentiation into TH1, TH2, and TH17 lineages.
- Required for establishment and maintenance of immunological memory for both B- and T-cells.
- Subcellular localization: predominantly nuclear, consistent with its role as a transcription factor.
- Expressed in germinal center T- and B-cells and primary immature dendritic cells.
