Biological Background: B7-H2/CD275 (ICOS Ligand) Function and Localization

  • ICOSLG encodes B7-H2 (CD275), a member of the immunoglobulin superfamily and the B7 family of immune regulatory proteins; alternative splicing generates multiple isoforms.
  • Functions as a ligand for the T-cell-specific receptor ICOS, providing a costimulatory signal for T-cell proliferation and cytokine secretion. Related references: PMID:11007762 PMID:11023515 PMID:30498080
  • Also induces B-cell proliferation and differentiation into plasma cells, contributing to adaptive immunity.
  • Mediates local tissue responses to inflammatory conditions and modulates secondary immune responses by costimulating memory T-cell function (By similarity).
  • In endothelial cells, required for proper neutrophil transmigration in response to chemoattractants such as CXCL8/IL8 and N-formyl-methionyl peptides. Related references: PMID:30498080
  • Expressed on the cell membrane of peripheral blood B-cells, monocytes, monocyte-derived dendritic cells, and widely in lymphoid and non-lymphoid tissues (brain, heart, kidney, liver, lung, pancreas, placenta, skeletal muscle, bone marrow, colon, ovary, prostate, testis, lymph nodes, leukocytes, spleen, thymus, tonsil).
  • Features immunoglobulin-like domains, disulfide bonds, and glycosylation; associated with keywords: adaptive immunity, B-cell activation, glycoprotein, signal, transmembrane.

Experimental Guidance and Technical Tips

  • The immunogen spans the extracellular domain (amino acids 19–256), suggesting the antibody likely recognizes the native conformation on live cells, making it suitable for flow cytometry.
  • As a direct conjugate, this antibody simplifies staining protocols by eliminating the need for a secondary antibody; titrate to determine the optimal concentration for your cell system.
  • Verify performance using human B-cell lines or activated monocytes as positive controls, and resting T cells as a negative control; stimulation with PMA/ionomycin or CD40L may upregulate B7-H2 expression.
  • Reported cross-reactivity is human; if working with other species, validate specificity before use.
  • For intracellular staining, consider that the epitope resides in the extracellular portion; appropriate permeabilization methods may be required if targeting internal pools.
