Biological Background: AXL Function and Localization
- AXL, encoded by the AXL (UFO) gene, is a receptor tyrosine kinase belonging to the TAM family (Tyro3, AXL, Mer) and is also known as ARK, UFO, JTK11, and Tyro7.
- AXL transduces extracellular signals by binding its ligand GAS6, regulating cell survival, proliferation, migration, and differentiation. Ligand binding induces dimerization and autophosphorylation, activating downstream AKT signaling via PI3-kinase and GRB2.
- AXL localizes at the cell membrane as a single-pass type I membrane protein.
- AXL inhibits Toll-like receptor (TLR)-mediated innate immune signaling, modulating the immune response.
- AXL serves as a host cell receptor for multiple viruses, including Lassa virus, lymphocytic choriomeningitis virus, Ebolavirus, and Zika virus. For Zika virus, AXL promotes entry in glial cells, Sertoli cells, and astrocytes and potentiates kinase activity to antagonize type I interferon signaling via a SOCS1-dependent mechanism.
