Biological Background: AXL Function and Localization

  • AXL, also known as UFO, ARK, JTK11, and Tyro7, is a receptor tyrosine kinase encoded by the AXL gene (GeneID 558).
  • It transduces signals by binding the ligand GAS6, regulating cell survival, proliferation, migration, and differentiation.
  • Upon ligand binding, AXL dimerizes, autophosphorylates, and activates multiple downstream signaling pathways, including PI3K-AKT, via phosphorylation of PIK3R1, GRB2, and others.
  • AXL is located on the cell membrane and is highly expressed in metastatic colon tumors, but weakly in normal colon tissue.
  • It plays a role in innate immunity by inhibiting Toll-like receptor (TLR)-mediated responses.
  • AXL serves as a host cell receptor for multiple viruses, including Lassa virus, lymphocytic choriomeningitis virus, and Ebola virus, likely via GAS6 bridging to phosphatidylserine on virions.
  • Zika virus utilizes AXL for entry into glial cells, Sertoli cells, and astrocytes, and potentiates AXL kinase activity to antagonize type I interferon signaling via SOCS1, promoting infection. Related references: PMID:28076778, PMID:29379210, PMID:31311882
  • Post-translational modifications include phosphorylation, glycosylation, disulfide bond formation, and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 33-449) of human AXL; this design is well-suited for detecting native surface receptor on live cells by flow cytometry.
  • Consider validating the antibody in a relevant human cell line with known AXL expression (e.g., metastatic colon cancer cells) and using proper isotype controls.
  • For flow cytometry, titrate the antibody to determine the optimal staining concentration and use a viability dye to exclude dead cells.
