Biological Background: GNAI3 Function and Localization
- GNAI3 is a member of the G(i) alpha subunit family, functioning as a molecular switch in GPCR signaling. It cycles between GDP-bound inactive and GTP-bound active states.

- In its active state, GNAI3 inhibits adenylate cyclase to reduce cAMP levels and stimulates receptor-regulated potassium channels. Related references: PMID:19478087 PMID:2535845
- GNAI3 plays a role in cell division by regulating the centrosomal localization of RGS14; the active GTP-bound form prevents RGS14 association with centrosomes. Related references: PMID:17635935

- For Western blot, GNAI3 is expected at ~41 kDa. Use appropriate loading controls and optimize antibody dilution.
- In immunofluorescence/immunocytochemistry, expect cytoplasmic, membrane, and possibly centrosomal staining. Permeabilization may be required for intracellular detection.
