Biological Background: Flotillin-2 Function and Localization
- Flotillin-2 (also known as ESA, ECS1, ESA1, ECS-1, M17S1) is an approximately 47 kDa membrane-associated protein encoded by the FLOT2 gene in humans.
- It functions as a scaffolding protein within caveolar membranes and is thought to participate in the formation of caveolae or caveolae-like vesicles.
- Flotillin-2 may also play a role in epidermal cell adhesion and contribute to the structural integrity and function of the epidermis.
- Subcellularly, it is found at the cell membrane, specifically in caveolae, as well as in endosomes and other membrane compartments.
- In skin, Flotillin-2 is expressed in the epidermis and epidermal appendages (but not the dermis), predominantly in suprabasal layers; it is also detected in melanoma and carcinoma cell lines, fibroblasts, and melanocytes.
- Based on its amino acid sequence and annotations, Flotillin-2 is subject to post-translational modifications such as myristoylation, palmitoylation, and phosphorylation, which likely influence its membrane anchoring and function.
Experimental Guidance and Technical Tips
- The immunogen was a recombinant protein spanning amino acids 239–428 of human FLOT2 (NP_004466.2). This region is well conserved, so the antibody may cross-react with Flotillin-2 from other species; verify in your system.
- For Western blot (WB) analysis, a specific band near 47 kDa is anticipated; always include appropriate positive and negative controls in your experimental design.
- In ELISA applications, consider using the recombinant immunogen as a positive control and optimize the antibody dilution for your specific assay format.
- Given the antibody’s reactivity with mouse Flotillin-2, it can be used in mouse-derived samples or xenograft models; however, validate performance in each tissue or cell type.
