Biological Background: CD14 Function and Localization
- CD14 is a monocyte differentiation antigen (My23 antigen, myeloid cell-specific leucine-rich glycoprotein) that serves as a coreceptor for bacterial lipopolysaccharide (LPS). Related references: PMID:1698311, PMID:23264655
- In concert with LBP, CD14 binds monomeric LPS and delivers it to the LY96/TLR4 complex, thereby mediating the innate immune response to bacterial LPS. Acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Related references: PMID:20133493, PMID:22265692, PMID:23264655, PMID:8612135
- Functions as a coreceptor for TLR2:TLR6 heterodimer in response to diacylated lipopeptides and for TLR2:TLR1 heterodimer in response to triacylated lipopeptides; these signaling clusters are targeted to the Golgi in a lipid-raft dependent pathway. Related references: PMID:16880211
- Binds electronegative LDL (LDL(-)) and mediates the cytokine release induced by LDL(-). Related references: PMID:23880187
- Subcellular localization includes the cell membrane, membrane rafts, and Golgi apparatus; a secreted form also exists.
- Tissue specificity: expressed strongly on the surface of monocytes and weakly on the surface of granulocytes; also expressed by most tissue macrophages.
- Keywords: GPI-anchor, leucine-rich repeat, glycoprotein, innate immunity, inflammatory response.
Experimental Guidance and Technical Tips
- The immunogen covers the extracellular domain (amino acids 20–352), suggesting the antibody likely recognizes an extracellular epitope suitable for staining live, non-permeabilized cells in flow cytometry.
- Consider direct immunofluorescence staining of human peripheral blood mononuclear cells (PBMCs) or cynomolgus leukocyte preparations; titrate the antibody to determine the optimal working concentration.
- Validate performance in the relevant sample system and include appropriate isotype controls and fluorescence compensation when using conjugate.
- Cross-reactivity with cynomolgus CD14 enables translational studies in non-human primate models.
