Biological Background: BGAT Function and Localization
- Belongs to the glycosyltransferase family and is the key enzyme determining the ABO blood group system. The protein exists in different allelic forms (A and B transferases) that catalyze the addition of N-acetyl-alpha-D-galactosamine (A antigen) or galactose (B antigen) to the H antigen precursor, while O alleles produce inactive protein.
- Encoded by the ABO gene, the transferase catalyzes the alpha1,3 linkage of GalNAc or Gal to the terminal galactose of all five types of naturally occurring H antigens, forming A and B blood group antigenic structures.
- Localized primarily to the Golgi stack membrane, where it functions in glycosylation; a secreted form has also been reported.
- Expressed at high levels in testis, with additional expression in pancreas, uterus, lung, and salivary gland.
- Classified as a metal-binding glycoprotein, requiring manganese as a cofactor for transferase activity.
- The protein is a single-pass type II membrane protein with a signal-anchor transmembrane helix, and undergoes proteolytic cleavage to generate a soluble form.

- The polyclonal nature may provide robust detection across species (human, mouse, rat). For Western blot, the predicted molecular weight is ~41 kDa; consider using whole cell lysates from blood cells or tissues known to express ABO transferase (e.g., testis, pancreas).
- For ELISA, validate the antibody with recombinant protein or cell lines expressing specific ABO antigens.
- Due to sequence similarity among ABO transferase alleles, the antibody may detect multiple variants; verify specificity in the experimental system.
