Biological Background: ATF5 Function and Localization
- ATF5, also known as ATFx, is a cAMP-dependent transcription factor that binds CRE, ARE, and AARE elements in target gene promoters.
- It critically regulates cell survival, proliferation, and differentiation in a cell-type-dependent manner. Related references: PMID:10373550, PMID:15358120, PMID:20654631
- Promotes cell proliferation and survival by inducing EGR1 synergistically with ELK1, and upon acetylation by EP300, binds ARE to activate BCL2 and EGR1. Related references: PMID:21791614
- In liver, cooperates with NR1I3 to activate CYP2B6 transcription, represses CRE-dependent transcription to block proliferation at G2/M, and modulates IL1B signaling via NLK-mediated C/EBP activation. Related references: PMID:18332083, PMID:18701499, PMID:22528486, PMID:24379400, PMID:25512613
- Functions as a cofactor of CEBPB to promote adipocyte differentiation. Related references: PMID:24216764
- Regulates centrosome dynamics by forming a 9-foci ring scaffold around the mother centriole. Related references: PMID:26213385
- Subcellular localization includes cytoplasm, nucleus, and centrosome; broadly expressed with highest levels in liver.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region in the C-terminal half of ATF5, which may recognize full-length and certain isoforms; validate in relevant sample systems.
- This antibody is validated for WB, IHC-P, and ELISA. For WB, an expected band at ~31 kDa; consider nuclear/cytoplasmic fractionation to enrich signal.
- Cross-reactivity with mouse and rat suggests a conserved epitope; verify in each species if used for non-human samples.
- For IHC-P, antigen retrieval and dilution optimization are recommended.
