Biological Background: PTBP1 Function and Localization
- PTBP1, also known as hnRNP I or 57 kDa RNA-binding protein, is a member of the heterogeneous nuclear ribonucleoprotein family.
- It plays a key role in pre-mRNA splicing by binding polypyrimidine tracts and regulating alternative splicing events, including exon skipping in its own pre-mRNA during muscle cell differentiation.
- PTBP1 represses the splicing of MAPT/Tau exon 10. Related references: PMID:15009664
- It binds to the polypyrimidine-rich controlling element (PCE) of CFTR and promotes exon skipping of CFTR exon 9, antagonizing TIA1. Related references: PMID:14966131
- PTBP1 regulates pyruvate kinase PKM splicing by repressing exon 9 inclusion, leading to production of the PKM M2 isoform. Related references: PMID:20010808
- Upon picornavirus infection, PTBP1 binds to the viral IRES and stimulates IRES-mediated translation. Related references: PMID:21518806
- The protein is predominantly localized in the nucleus and undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an N-terminal sequence (aa 1-100) of human PTBP1; confirm antibody reactivity against full-length protein in your experimental system.
- For Western blotting, a band around 60 kDa is expected; optimize loading and antibody dilution for your samples.
- In IHC-P, antigen retrieval may be required; validate with appropriate positive control tissues.
- For IF/ICC, consider using 4% PFA fixation and permeabilization with Triton X-100.
- ELISA applications may require optimization of coating antigen and antibody concentration; test under your assay conditions.
- Cross-reactivity with human and mouse is reported; verify reactivity in other species if needed.
