Biological Background: PTBP1 Function and Localization

  • PTBP1, also known as hnRNP I or 57 kDa RNA-binding protein, is a member of the heterogeneous nuclear ribonucleoprotein family.
  • It plays a key role in pre-mRNA splicing by binding polypyrimidine tracts and regulating alternative splicing events, including exon skipping in its own pre-mRNA during muscle cell differentiation.
  • PTBP1 represses the splicing of MAPT/Tau exon 10. Related references: PMID:15009664
  • It binds to the polypyrimidine-rich controlling element (PCE) of CFTR and promotes exon skipping of CFTR exon 9, antagonizing TIA1. Related references: PMID:14966131
