Biological Background: PSMA7 Function and Localization
- PSMA7 encodes proteasome subunit alpha type-7, also known as proteasome subunit RC6-1, XAPC7, or alpha-4. UniProt: O14818.
- Core component of the 20S proteasome, a barrel-shaped complex that performs proteolytic degradation of intracellular proteins. Associates with regulatory particles to form 26S proteasome (ATP-dependent ubiquitinated protein degradation), or with PA200/PA28 for ubiquitin-independent degradation (e.g., spermatogenesis, MHC class I antigen processing).
- Plays roles beyond proteolysis: inhibits HIF-1A transactivation under normoxic and hypoxic conditions; interaction with EMAP2 enhances HIF-1A degradation.
- Mediates hepatitis C virus internal ribosome entry site (IRES)-mediated translation and promotes nuclear translocation of androgen receptor, enhancing androgen-mediated transactivation.
- Negatively regulates innate immune response by promoting MAVS degradation.
- Localizes to both cytoplasm and nucleus.
- Post-translational modifications include acetylation, phosphorylation, and glycosylation; alternative splicing produces isoforms.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human PSMA7 (residues 1–248), which covers the entire alpha-4 subunit. Expect detection of the ~28 kDa band in western blot.
- For IHC-P and IF/ICC, note that PSMA7 localizes to both cytoplasm and nucleus; ensure appropriate fixation and permeabilization conditions for nuclear antigens.
- Cross-reactivity with mouse and rat has been reported; verify in your specific sample matrix.
- Suitable for ELISA applications; consider using with recombinant PSMA7 or cell lysates.
