Biological Background: PRMT1 Function and Localization

  • PRMT1 is a protein arginine N-methyltransferase encoded by the PRMT1 gene, which undergoes alternative splicing and is also known as HRMT1L2, IR1B4, HCP1, or ANM1. It is a member of the protein arginine methyltransferase family and functions as the main enzyme catalyzing arginine monomethylation and asymmetric dimethylation on target proteins.
  • Core methyltransferase function: Methylates a wide range of substrates, including histones (H2, H3, H4) and non-histone proteins (ESR1, FMR1, HNRNPA1, ILF3, HNRNPD, NFATC2IP, SUPT5H, TAF15, EWS, HABP4, SERBP1, RBM15, FOXO1, CHTOP, MAP3K5/ASK1, MICU1, NPRL2), playing central roles in epigenetic regulation, signal transduction, and RNA processing. It specifically methylates histone H4 at Arg-3 (H4R3me1 and H4R3me2a), a mark associated with transcriptional activation, and also methylates H4R3 in genes involved in glioblastomagenesis in a CHTOP- and/or TET1-dependent manner. Related references: PMID:10749851 PMID:15741314 PMID:16879614 PMID:25284789
  • DNA damage response: Methylates MRE11 and TP53BP1, promoting the DNA damage response and genome stability. Related references: PMID:15741314 PMID:16294045 PMID:29651020
  • Regulation of signaling proteins: Methylates FOXO1 (Arg-248/Arg-250) to retain it in the nucleus and enhance its transcriptional activity; methylates CHTOP to facilitate its 5hmC-binding activity; methylates MAP3K5/ASK1 (Arg-78/Arg-80) to inhibit JNK activation; methylates NPRL2 (Arg-78) to regulate mTORC1 signaling; methylates DSP to promote its phosphorylation and recruitment to cell junctions. Related references: PMID:18951090 PMID:25284789 PMID:22095282 PMID:38006878 PMID:25733715
  • RBM15 methylation: Methylates RBM15, promoting its ubiquitination and subsequent degradation, potentially influencing RNA processing and translational regulation. Related references: PMID:26575292
  • Subcellular localization: Found predominantly in the nucleus and nucleoplasm, but also detected in the cytoplasm, cytosol, and lysosome membrane, depending on the cellular context and binding partners.
  • Tissue specificity and cancer relevance: Widely expressed in human tissues, with notably strong expression in colorectal cancer cells and tissues compared to normal colon samples, suggesting a role in oncogenesis. Related references: PMID:11097842 PMID:28040436
  • Post-translational modifications: PRMT1 is subject to acetylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation, as identified by proteomics studies. These modifications may regulate its enzymatic activity, stability, or subcellular distribution.

Experimental Guidance and Technical Tips

  • Immunogen design: The immunogen corresponds to the C-terminal region (amino acids 270–371) of human PRMT1, which is highly conserved across human, mouse, and rat, consistent with the observed cross-reactivity. This region includes part of the catalytic domain and may generate antibodies that recognize the active conformation.
  • Application suitability: The antibody is qualified for Western blotting (WB; 42 kDa band expected), immunohistochemistry (paraffin), immunofluorescence/immunocytochemistry, and ELISA. For quantitative detection, ELISA may be considered; for spatial resolution, IF/ICC or IHC-P are options.
  • Technical considerations: As a polyclonal antibody, batch-to-batch consistency should be verified. End-users should validate the antibody in their specific sample context, using appropriate positive/negative controls and titration experiments.
