Biological Background: PPIL2 Function and Localization
- PPIL2, also known as Cyp-60 or CYC4, is a RING-type E3 ubiquitin-protein ligase that mediates 'Lys-48'-linked polyubiquitination, targeting substrates for proteasomal degradation. Related references: PMID:11435423
- It may also function as a chaperone, facilitating the transport of proteins like BSG/Basigin to the cell membrane. Related references: PMID:15946952
- PPIL2 is a probable inactive peptidyl-prolyl cis-trans isomerase (PPIase), with no rotamase activity demonstrated. Related references: PMID:20676357
- As a component of the minor spliceosome, PPIL2 is involved in the splicing of U12-type introns in pre-mRNAs (Probable).
- The protein is predominantly localized in the nucleus.
- PPIL2 shows highest expression in thymus, pancreas, and testis, with strong protein expression also observed in lymph nodes, thyroid, testis, stomach, pancreas, kidney, endometrium, and colon. It is highly expressed in most lymphomas and melanomas.

- The immunogen corresponds to amino acids 1-260 of human PPIL2 (NP_680481.1). This N-terminal region may be suitable for detecting full-length PPIL2 and potential isoforms, but cross-reactivity with other cyclophilin family members should be excluded by sequence alignment.
- For Western blot, the predicted molecular weight of PPIL2 is approximately 58.8 kDa; a band around 59 kDa is expected under reducing conditions. Validate the signal in a relevant sample system, such as thymus or testis lysates, where expression is highest.
- In ELISA, consider using purified recombinant PPIL2 as a standard and titrate the antibody to determine optimal working concentrations.
