Biological Background: POLR2A Function and Localization
- DNA-directed RNA polymerase II subunit RPB1 (POLR2A) is the largest and catalytic subunit of RNA polymerase II, essential for transcribing protein-coding genes and numerous non-coding RNAs. Related references: PMID:23748380 PMID:27193682 PMID:30190596 PMID:28108474
- The polymerase catalyzes the template-directed addition of ribonucleotides, with its active site coordinated by magnesium ions; it also possesses a 3'→5' exonuclease proofreading activity. Related references: PMID:8381534 PMID:9852112
- A unique C-terminal domain (CTD) containing 52 heptapeptide repeats acts as a regulatory hub, with phosphorylation at Ser5 facilitating promoter escape and Ser2 promoting elongation and pre-mRNA processing. Related references: PMID:24207025 PMID:26124092
- Beyond DNA-dependent transcription, POLR2A can function as an RNA-dependent RNA polymerase, extending non-coding RNAs using an internal template, potentially influencing gene silencing.
- Localized primarily in the nucleus, with presence in the cytoplasm and on chromosomes; post-translational modifications include acetylation, methylation, ubiquitination, and phosphorylation.
- Mutations in POLR2A are associated with neurodevelopmental disorders, and the protein is targeted by viruses such as Hepatitis delta virus for replication of its genome.
Experimental Guidance and Technical Tips
- For Western blot, due to the large protein size (217 kDa), use a 6-8% polyacrylamide gel and ensure efficient transfer (e.g., wet transfer with 10% methanol). A positive control such as HeLa or HEK293 cell lysate may be considered.
- For IF/ICC and IHC-P, fixation and antigen retrieval conditions should be optimized; permeabilization with Triton X-100 may be necessary for intracellular nuclear targets.
- For ChIP, the N-terminal epitope location suggests suitability for detecting native POLR2A, but validation with positive and negative target genes is recommended.
- As a polyclonal antibody, lot-to-lot consistency is expected but should be verified in the intended application.
