Biological Background: PNPLA3 Function and Localization
- PNPLA3 catalyzes the CoA-dependent acylation of 1-acyl-sn-glycerol 3-phosphate (LPA) to generate phosphatidic acid (PA), a critical precursor in triglyceride and phospholipid synthesis. It preferentially utilizes long-chain fatty acyl-CoAs (≥C16) such as arachidonoyl-CoA and oleoyl-CoA (PubMed:22560221). Related references: PMID:22560221

- The predicted molecular weight is ~53 kDa; however, glycosylation and other modifications may cause band shifts in Western blot, so use appropriate positive controls such as human liver or HepG2 lysates.
- For ELISA, optimize coating buffer and detection antibody concentrations using recombinant PNPLA3 as a standard.
- Cross-reactivity with mouse and rat is indicated; validate by comparing sequences in the immunogen region to ensure epitope conservation.
