Biological Background: PNN Function and Localization

  • Pinin (PNN) is a nuclear and cell adhesion-related phosphoprotein, also known as DRS, DRSP, SDK3, or memA.
  • Functions as a transcriptional activator by binding to the E-box 1 core sequence (5'CAGGTG-3') of the E-cadherin promoter, capable of reversing CTBP1-mediated transcription repression.
  • Serves as an auxiliary component of the exon junction complex (EJC), deposited at splice junctions on mRNAs, and participates in the regulation of alternative pre-mRNA splicing.
  • Associates with spliced mRNA within 60 nucleotides upstream of the 5'-splice site.
  • Component of the PSAP complex, which binds RNA in a sequence-independent manner and is proposed to be recruited to the EJC to regulate specific intron excision.
  • Plays a role in establishing and maintaining epithelial cell-cell adhesion.
  • Acts as a potential tumor suppressor for renal cell carcinoma.
  • Localizes to nuclear speckles and desmosomes at cell junctions.
  • Expressed in a wide range of tissues and strongly in melanoma metastasis lesions and advanced primary tumors.
  • Post-translationally modified by acetylation, methylation, phosphorylation, and ubiquitination.

Experimental Guidance and Technical Tips

  • For Western blot (WB), consider using whole cell lysates from human or rat cell lines, and validate under reducing conditions. The predicted molecular weight is 82 kDa, but post‑translational modifications may cause migration shifts.
  • Since the immunogen lies within the N‑terminal 1–100 amino acids, the antibody may recognize multiple isoforms or processed forms; this can be assessed by comparing band patterns in relevant tissues.
  • For ELISA, a standard indirect protocol with the immunogen peptide as coating antigen is advised; detection of native protein can be validated using cell lysates.
  • Given cross‑reactivity with rat, this antibody may be suitable for rodent models, but optimal working dilutions and fixation/permeabilization conditions should be determined empirically.
