Biological Background: PIWIL4 Function and Localization
- PIWIL4 (also designated HIWI2 or MIWI2) is a Piwi-like protein and member of the Argonaute family, central to piRNA-mediated gene silencing.
- Essential for spermatogenesis by repressing transposable elements and maintaining germline integrity via the piRNA pathway (By similarity).
- Functions in the nucleus to silence active transposons by engaging nascent transposable element transcripts and directing DNA methylation and repression (By similarity).
- Lacks endonuclease activity, acting as a 'slicer-incompetent' component in the piRNA amplification (ping-pong) loop, loading cleaved piRNAs from PIWIL2 to target genomic transposon loci (By similarity).
