Biological Background: PIK3CG Function and Localization
- PIK3CG encodes the p110γ catalytic subunit of class I phosphoinositide 3-kinase (PI3K), also known as PI3Kγ, a lipid kinase that phosphorylates PtdIns(4,5)P2 to generate PIP3.
- PIP3 serves as a membrane anchor for PH domain-containing proteins such as AKT1 and PDPK1, triggering signaling pathways that regulate cell growth, survival, proliferation, and motility.
- p110γ links G protein-coupled receptor (GPCR) activation to PIP3 production, playing a crucial role in immune, inflammatory, and allergic responses.
- The protein is critical for leukocyte chemotaxis and migration by controlling PIP3 spatial accumulation, F-actin organization, and integrin-based adhesion at the leading edge.
- PIK3CG also possesses serine/threonine protein kinase activity, which together with its lipid kinase activity is required for GPCR endocytosis and regulation of cardiac contractility.
- It is expressed in pancreas, skeletal muscle, liver, and heart, and localizes to the cytoplasm and cell membrane.
- The protein is a negative regulator of cardiac contractility and contributes to pathological cardiac hypertrophy through kinase-dependent and scaffolding functions.
- Post-translational modifications and scaffolding interactions enable PIK3CG to assemble signaling complexes involved in angiogenesis, platelet aggregation, and neutrophil responses.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1-310) of human PIK3CG; this may allow detection of full-length and potentially truncated forms.
- For Western blot, expect a band near 126 kDa; validate in human tissues or cell lines known to express PIK3CG, such as leukocytes, heart, or skeletal muscle.
- In ELISA, consider optimizing the coating antigen and antibody dilution using recombinant protein or cell lysates.
- As a polyclonal antibody, performance may vary between lots; re-validate each new lot in the relevant assay system.
- Cross-reactivity with species other than human has not been characterized; for non-human samples, perform sequence alignment and experimental validation.
