  • PIGR (Polymeric Immunoglobulin Receptor) is a single-pass type I membrane protein that mediates the selective transcytosis of polymeric IgA and IgM across mucosal epithelial cells.
  • Upon binding polymeric immunoglobulins at the basolateral surface, the receptor-ligand complex is transported across the cell to the apical surface, where proteolytic cleavage releases the extracellular portion as the secretory component.
  • Through its N-linked glycans, the secretory component anchors secretory IgA (sIgA) molecules to the mucosal lining, helping to neutralize extracellular pathogens.
  • In its free, unliganded form, PIGR may act as a non-specific microbial scavenger, preventing pathogen adherence to epithelial cells.
  • PIGR is localized at the cell membrane and is also secreted after cleavage of its extracellular domain.
  • The protein contains immunoglobulin-like domains, disulfide bonds, and is subject to N-glycosylation and phosphorylation, which are critical for its stability and function.
