Biological Background: PIAS1 Function and Localization
- PIAS1, also named GBP, ZMIZ3, DDXBP1, or GU/RH-II, is an E3 SUMO-protein ligase and SUMO-tethering factor that catalyzes the covalent attachment of SUMO to substrates such as CEBPB, MRE11, PML, and ZNF76. Related references: PMID:11583632 PMID:11867732 PMID:14500712
- It functions as a transcriptional coregulator in multiple signaling pathways, including the STAT, p53, and steroid hormone pathways, where it can either activate or repress gene expression depending on the biological context. Related references: PMID:11583632 PMID:11867732
- In vitro, PIAS1 binds A/T-rich DNA, suggesting a direct role in chromatin targeting or transcriptional regulation. Related references: PMID:15133049
- PIAS1 mediates sumoylation of MRE11, which stabilizes MRE11 on chromatin during DNA end resection, linking it to DNA damage repair. Related references: PMID:36050397
- It enhances the sumoylation of the metastasis-associated protein MTA1 and may participate in its paralog-selective sumoylation. Related references: PMID:21965678
- PIAS1 restricts lytic replication of Epstein-Barr virus (EBV) by inhibiting transcription factors required for lytic gene expression, though the virus can use caspases to antagonize this restriction. Related references: PMID:29262325
- Subcellularly, PIAS1 localizes predominantly to the nucleus, including nuclear speckles and PML bodies, and is also found associated with the cytoskeleton.
- PIAS1 is ubiquitously expressed but shows highest expression in testis, as indicated by tissue profiling.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 1–140) of human PIAS1; consider sequence alignment with mouse and rat orthologs to assess cross-reactivity for your specific experiments.
- For Western blot (WB), a band at approximately 72 kDa is expected. Validate the antibody in your cell or tissue lysate system, as post-translational modifications or isoforms may affect migration.
- In ELISA applications, consider using a recombinant protein covering the immunogen region as a positive control to confirm binding specificity.
- As a polyclonal antibody, lot-to-lot consistency may vary; it is recommended to include appropriate positive and negative controls and to optimize the working dilution for each new batch.
- Since PIAS1 is predominantly nuclear, using nuclear-enriched lysates for WB may improve detection sensitivity.
- For quantitative WB analysis, normalize using total protein staining (e.g., stain-free technology) or a validated housekeeping protein to ensure accurate loading comparisons.
